以甜瓜(Cucumis melo)品种‘新银辉’为材料,采取根灌法,以水杨酸(10μmol/L)处理甜瓜幼苗,通过Real-Time PCR对叶片中部分自毒作用相关基因的表达量进行定量分析,以探究外源水杨酸诱导对甜瓜自毒作用的影响。结果显示,经外源水杨酸诱导后:(1)在催化苯丙烷类代谢途径的3个初始反应的蛋白酶编码基因中,PAL和C4H随处理后时间增加,呈逐渐上调趋势,但PAL比C4H更快响应胁迫,而4CL转录水平没有显著变化;(2)在类黄酮生物合成的3个关键酶的基因中,CHI、F3H均显示出不同程度的下调,CHS于处理后1 d略微上调,然后恢复至处理前水平;(3)两个转录因子基因WD40、R2R3-MYB分别在不同处理时间后显著下调;(4)WD40与CHI的表达相关性较高。据上述结果 ,水杨酸诱导后:(Ⅰ)CHI表达可能与转录因子WD40蛋白相关;(Ⅱ)WD40、R2R3-MYB与PAL、C4H、4CL、CHS间不存在简单、明确的调控关系;(Ⅲ)柚皮苷、表儿茶素、芦丁等甜瓜主要化感物质合成减少。  相似文献

Consumption of whole-wheat based products is encouraged due to their important nutritional elements that benefit human health. However, the use of whole-wheat flour is limited because of the poor processing and end-product quality. Bran was postulated as the major problem in whole wheat breadmaking. In this study, four major bran components including lipids, extractable phenolics (EP), hydrolysable phenolics (HP), and fiber were evaluated for their specific functionality in flour, dough and bread baking. The experiment was done by reconstitution approach using the 24 factorial experimental layout. Fiber was identified as a main component to have highly significant (P < 0.05) and negative influence on most breadmaking characteristics. Although HP had positive effect on farinograph stability, it was identified as another main factor that negatively impacted the oven spring and bread loaf volume. Bran oil and EP seemed to be detrimental to most breadmaking characteristics. Overall, statistical analysis indicates that influence of the four bran components are highly complex. The bran components demonstrate multi-way interactions in regards to their influence on dough and bread-making characteristics. Particularly, Fiber appeared to have a high degree of interaction with other bran components and notably influenced the functionality of those components in whole wheat bread-making.  相似文献

采用氨基酸自动分析仪测定分析大红袍、水仙、肉桂、雀舌、金毛猴和矮脚乌龙等6个品种武夷岩茶的游离氨基酸组分,结果表明:6个品种武夷岩茶均含有17种氨基酸,氨基酸含量品种间差异显著,武夷岩茶的游离氨基酸总量为75.82~107.12 mg/g;其中含量较高的为谷氨酸、天冬氨酸和亮氨酸。武夷岩茶的氨基酸中含有各种人体必需氨基酸,且比例均衡;必需氨基酸总量占30.47%~40.46%,药用氨基酸含量丰富,占氨基酸总量的61.29%~63.90%;呈鲜味的氨基酸含量也占氨基酸总量的23.80%~28.63%,因此,武夷岩茶具有较高的营养价值和保健功能。  相似文献

利用高效液相色谱法(HPLC)检测没食子酸(gallic acid,简称GA)在水稻叶片上的残留时间,并在检测水稻叶片上残留量的同时接种病原菌,调查接种后水稻细菌性条斑病的发病情况,以确定GA在水稻叶片上对水稻细菌性条斑病防治作用的持效期。研究结果表明,GA不具有内渗作用,只残留在水稻叶片的表面。分别用100、200和400 mg/L的浓度处理叶片后,GA在叶片表面的残留时间分别为16、24和28 d。200 mg/L浓度处理叶片后,GA对水稻细菌性条斑病防治作用的持效期为16 d。研究结果可为有效使用GA防治水稻细菌性条斑病奠定基础。  相似文献

研究红茶提取物对高尿酸血症小鼠血尿酸的影响。将36只雄性KM小鼠随机分为空白组、模型组、红茶提取物低、中、高剂量组及别嘌呤醇组。空白组和给茶组连续1周分别灌胃生理盐水和红茶提取物,给茶组第7天造模后1 h给茶;模型组在第7天腹腔注射氧嗪酸钾并灌胃酵母膏造模。测定结果显示:与模型组相比,各给茶组血尿酸(UA)水平均降低;与模型组相比,给茶组血尿素氮(BUN)水平均降低,其中、高剂量给茶组BUN水平显著降低(P0.05),高剂量组差异极显著(P0.01);各给茶组血肌酐(Cr)值与模型组相比极显著下降(P0.001)。高剂量组黄嘌呤氧化酶(XOD)活性较模型组显著降低(P0.05),低、中剂量组有一定的抑制作用,但无显著差异。研究表明,红茶提取物对氧嗪酸钾和酵母膏导致的小鼠高尿酸血症有明显的改善作用。  相似文献

The effects of a limited grazing period on the performance, behaviour and milk composition of high-yielding dairy cows were examined. A total of 56 Holstein cows yielding 44.7 ± 0.42 kg/day were allocated to one of four treatments in one of two, 4-week periods. Treatments were as follows: control (C)—cows housed and offered TMR ad libitum; early grazing (EG)—cows grazed for 6 hr after morning milking then housed; delayed grazing (DG)—cows returned to housing for 1 hr after morning milking followed by grazing for 6 hr, then housed; restricted TMR (RT)—cows grazed for 6 hr after morning milking, then housed and fed TMR at 75% of ad libitum. Intake of TMR was highest in cows receiving C, intermediate in EG and DG, and lowest in RT at 26.9, 23.6, 24.7 and 20.3 kg DM/day respectively. Pasture intake was similar in cows receiving EG or DG, but was higher in RT at 2.4, 2.0 and 3.5 kg DM/day respectively. Milk yield was similar between cows receiving C, EG or DG, but lowest in RT at 45.7, 44.2, 44.9 and 41.7 kg/cow, respectively, while milk fat content of C18:3 n-3 was increased by grazing. Cows in C spent more than 55 min/day longer lying and had three additional lying bouts/day, while lying bouts were shorter than for cows receiving EG, RT or DG. It is concluded that high-yielding cows can be grazed for 6 hr/day with little impact on performance, provided TMR is available ad libitum when housed.  相似文献

In this article, through the combination of nucleic acid probes and immune chromatography, a simple, sensitive and specific detection system——nucleic acid lateral flow immunoassay (NALFIA) for amplifing foot-and-mouth disease virus (FMDV) 3D RT-PCR products was established.An ultrasensitive nucleic acid biosensor (NAB) based on streptavidin-labeled gold nanoparticles dual labels and lateral flow strip biosensor (LFSB) were used in this system.The biotinylated goat anti-rabbit IgG was marked to the NC membrane as the alleged strip and the anti-digoxin antibody was labeled to the NC membrane to capture the digoxin probe.After assemblying gold-labeled strip and detecting RT-PCR products, the detection limit of NALFIA was 0.3×10-3 to 3×10-3 μg/μL.The NALFIA was compared with agar gel electrophoresis analysis, the results showed that the sensitivity of NALFIA was higher than agar gel electrophoresis.There was an excellent agreement between the two methods.NALFIA was a method with high sensitive, low cost and short time.In conclusion, this method provided a good alternative to detect FMDV.  相似文献

Plant leucine-rich repeat (LRR) domain-containing proteins are known to play important roles in signaling transduction and defense responses. In sorghum, SbLRR2 is pathogen-inducible gene encoding a simple extracellular LRR protein. Here, we demonstrated an earlier and stronger expression of SbLRR2 in a sorghum resistant genotype in comparison to a susceptible genotype following inoculation with the anthracnose pathogen (Colletotrichum sublineolum). In addition, SbLRR2 expression was found to be induced strongly by methyl-jasmonate treatment. Functional analysis was performed in SbLRR2 over-expression (OE) Arabidopsis plants, which showed enhanced resistance against the necrotrophic pathogens Botrytis cinerea and Alternaria brassicicola. In addition, the OE lines were found to have elevated expression of several jasmonate acid (JA)-associated genes and higher endogenous JA contents. Hence, the SbLRR2-mediated defense responses in transgenic Arabidopsis are likely to be dependent on JA-signaling through increased JA production. On the other hand, the OE lines remained susceptible to Pseudomonas syringae pv. tomato like the wild type plants. Consistently, there was no up-regulation of salicylic acid (SA) defense marker gene expression or SA levels in the OE lines. Our results suggested that SbLRR2 is potentially useful for enhancing resistance against necrotrophic pathogens in transgenic dicot crops.  相似文献

为了筛选出有利于荔枝贮藏的复合保鲜剂,以‘井岗红糯’荔枝果实为试验材料,通过正交试验研究了不同浓度的油菜素内酯(brassinolide, BL)和曲酸(kojic acid, KA)配比对采后荔枝果实的保鲜效果。结果表明,在25 ℃贮藏条件下,对荔枝果实的最佳保鲜配方为:油菜素内酯40 μmol/L、曲酸35 mmol/L,浸泡时间为3 min,该复合保鲜剂配方能较好地抑制荔枝果实褐变和腐烂,降低果皮相对电导率、果皮pH和果皮丙二醛(malonaldehyde, MDA)含量,延缓果肉总可溶性固形物(total soluble solids, TSS)和维生素C(vitamin C, VC)含量的下降,维持较高的果皮色度L *值、a *值、C *值和花色苷含量,抑制了多酚氧化酶(polyphenol oxidase, PPO)、过氧化物酶(peroxidase, POD)及漆酶(laccase, Lac)的活性。  相似文献

针对目前测定甜玉米还原糖和蔗糖含量的3,5-二硝基水杨酸法和酶法进行比较研究,分析两者在育种中的应用。结果显示:酶法的测量准确度比3,5-二硝基水杨酸法高,且其稳定性和重现性都优于3,5-二硝基水杨酸法。通过两种方法对10个甜玉米杂交种种子的还原糖和蔗糖含量的测定,证实了两种方法在甜玉米还原糖和蔗糖含量的检测上是一致的。从育种实践考虑,酶法适于样品数较小的检测,3,5-二硝基水杨酸法更适合于大批量样品的检测。  相似文献
